Background: The technology related to the establishment of primary tumor cell cultures of solid tumors, including glioblastoma, which is increasingly important for oncology research and practice. However, the processing of fresh tumor specimens for the formation of primary culture on the day of collection logistically difficult surgery. Feasibility cryopreservation of decent specimens of glioblastoma, allowing for major cultural establishments weeks to months after surgical tumor collection and freezing, shown by Mullins et al. in 2013, with a success rate of 59% which is not significantly lower than that achieved with fresh tumor tissue. However, research is targeting the optimization of glioblastoma cryopreservation protocol for the establishment of a culture worthy of the primary tumor has been limited.
Objective: The objective of this study was to optimize the cryopreservation method for cryobanking decent glioblastoma and to determine whether the two-dimensional (2D) or three-dimensional (3D) culture conditions that favor the growth of glioblastoma after thawing frozen tumor specimens. Methods: Part of eight human glioblastoma specimens cryopreserved by four different protocols differ in enzymatic digestion time (before or after cryopreservation), and in the cryopreservation media types (CryoStor CS10 or 10% dimethyl sulfoxide and 90% fetal calf serum).
After 1 month, the frozen tissue is thawed, enzymatically digested, if not digested in advance, and are used to culture primary tumor initiation 2D or 3D to determine eligibility. Results: Among the cryopreservation were tested and culture protocol, a combination of the most efficient of the cryopreservation and culture associated with the use of CryoStor CS10 cryopreservation medium, enzymatic digestion before it freezes, and 2D culturing after thawing the cultural level of success than 8 out of 8 cases (100% ). two-dimensional culture is generally more efficient to support the growth of tumor cells after thawing of 3D culture. Conclusion: The development of supporting worthy glioblastoma study cryopreservation methods based evidence for use in glioblastoma biobanking and research.
[Construction of RNA interference (RNAi) gene DEK lentiviral expression vector and its effect on the biological behavior of liver cancer cells]
Objective: To construct RNA interference (RNAi) gene DEK lentiviral expression vector, and to explore its effect on biological behavior of liver cancer cells. Methods: Double-stranded DNA oligo amplified and synthesized according to the sequence of the gene disruption DEK by RNAi technology. Disturb small RNA expression vector cloned pLKO.1 after enzymatic digestion. Recombinant lentiviral-sh pLKO.1 hDEK built, and then the virus supernatant was collected, packaged and infected by 293T cells. Real-time reverse transcription PCR (RT-PCR) and Western blot is used to detect the expression of DEK on human liver cancer cells Bel-7402, Hu-7, SmMC-7721 and HepG2, and DEK efficiency knockdown in each group of lentivirus-infected cells.

T-test was used to compare the mean between two groups, and one-way ANOVA was used to compare the groups. Results: enzymatic digestion and DNA sequencing results confirm that recombinant lentiviral vector pLKO.1-sh-sh hDEK1 and pLKO.1 hDEK3 successfully constructed. RT-PCR and Western blot results showed that DEK expression in human liver cancer cells BEL-7402 and higher Huh7 cells, and pLKO.1-sh hDEK3 more effective in inhibiting the DEK gene expression (P <0.05).

Reactive Oxygen Species Induces Differentiation of endothelial Liver Cancer Stem-Like Cells Sphere through the activation of Akt / IKK Signaling Pathway
Cancer stem cells (CSC) of a variety of cancers can be transdifferentiate into endothelial cells and further form functional blood vessels, suggesting possible resistance mechanism other antiangiogenic agents. However, it remains unclear whether CSC of hepatocellular carcinoma has the ability to differentiate into endothelial cells, and the resulting resistance to antiangiogenic therapies targeting VEGF. Reactive oxygen species (ROS) are involved in self-renewal and differentiation of CSC, however, their role in endothelial differentiation of CSC has been poorly understood. In this study, we found that the cancer stem-like ball of cells enriched from cell line hepatocellular carcinoma human Hep G2 can differentiate into endothelial cells morphologic and functional, and this process could be blocked by Akt1 / 2 kinase inhibitor and IKK-β inhibitor BAY 11 -7082 but not with Bevacizumab, VEGFA-binding antibody, and dAPT, γ-Secretase inhibitor.
Both hydrogen peroxide and BSO (inhibitor of GSH biosynthesis) induce differentiation of cancer stem-like cells ball into endothelial cells, which can be canceled by the antioxidant N-Acetyl-L-cysteine (NAC). We also found that hydrogen peroxide or BSO induces phosphorylation of Akt and IKK scope of differentiated endothelial cells.
Thus, both Akt1 / 2 kinase inhibitor BAY 11-7082 and inhibits hydrogen peroxide and BSO-mediated endothelial differentiation of cancer stem-like cells of the ball. Collectively, these results indicate that the cancer stem-like ball of Hep G2 cells can differentiate into endothelial cells both morphologically and functionally, and this process is independent of VEGF and NOTCH signaling but depends on the activation of Akt and IKK. ROS promotes endothelial differentiation of cancer stem-like cells through activation of Akt scope / IKK signaling pathways. Therefore, our study suggests a new mechanism of resistance to conventional antiangiogenic therapy and may provide potential therapeutic targets for the treatment of liver cancer.
