More recently, researchers have focused on the role of gut microbiota on human health and reported a two-way relationship between the gut microbiota and the brain, referred to as axis-brain-gut microbiota (MGBA). In this context, the development platform organ-on-a-chip recapitulation of the main players of MGBA will assist in the investigation of the biochemical mechanisms involved. In this work, we focus on developing new network models, 3D hydrogel-based brain-like compartment which will be held in the brain of the platform.
We previously cultured primary mouse microglia cells, cortical neurons and astrocytes independently, once embedded or covered by a two millimeter layer of collagen-based hydrogels are selected. We evaluate the metabolic activity of cells up to 21 days, cell morphology, spatial distribution and synapse formation. Then, we exploited the best performance culture conditions and develop brain-like networks more complex models based on co-cultures of cortical neurons and glial cells under physiological conditions. The results showed that our brain network model-based 3D hydrogel suitable for in vitro recapitulation of the key biochemical parameters of brain tissue.

Therefore, understanding the regulatory mechanisms in HCC LCSCs is very important. In this review, we provide an update on recent advances in experimental research on non-coding RNAs (ncRNAs), oncogenes and Oncoproteins.
All articles discuss the crosstalk between the different ncRNAs, oncogenes and Oncoproteins, as well as upstream and downstream products they are targeting LCSCs. In this review, we summarize the three paths, the Wnt / β-catenin signaling pathway, phosphatidylinositol 3-kinase (PI3K) / protein kinase B (Akt) signaling pathway, and interleukin 6 / Janus kinase 2 / signal transducer and activator of transcription 3 ( IL6 / JAK2 / STAT3) signaling pathways, and genes they target, c-Myc. In addition, we conclude that the octamer 4 (OCT4) and Nanog are two important functional genes that play important roles in the regulation of LCSCs and prognosis of HCC.
Effect of Diamond Particle Fluorescent FDP-NV-800Nm on the biochemical function of Essential Human Primary Cells Human Umbilical Vein and Hepatic Cells, HepG-2 in vitro (Part VI): Acute Study of Biocompatibility
Background: Recently, we reported the safety and biocompatibility of diamond particles neon, FDP-NV-Z-800Nm (FDP-NV) was injected intravenously into mice, where no morbidity and mortality were recorded over a period of 3 month. Acute effects of particle FDP-NV-800Nm in cultured human endothelial cells and the heart remains unexplored.
Objective: In this study, we aimed to explore the function of cells and biochemical select in cultured human cells umbilical endothelial (HUVEC) and human liver cancer cells (HepG-2) exposed FDP-NV-800 in vitro at levels of exposure in the pharmacokinetics (Cmax and nadir) were previously reported in vivo.
Methods: Various biochemical function of cells and monitored, which cumulatively can provide insight into some vital cellular functions. Cell proliferation and migration was assessed by quantitative microscopy. mitochondrial metabolic function was tested by MTT assay, and cytosolic esterase activity was studied by assay Calcein AM. Chaperons (CHOP), BIP and apoptosis (caspase-3 activation) monitored using Western blot (WB). MAPK Erk1 / 2 signaling was assessed by detection of the phosphorylated form of the protein (P-Erk 1/2) and translocated to the cell nucleus.
Results: At all concentrations tested (0.001-0.1mg / mL), FDP-NV does not affect any of the biomarker integrity HepG2 cells. On the contrary, the proliferation of HUVEC effected at the highest concentration tested (0.1mg / mL, Cmax). HUVEC exposure to (0.01 mg / mL) FDP-NV had mild-moderate effect on cell proliferation as shown in the MTT test and was not present when the proliferation was assessed by direct cell counting or by using the Calcein AM assay. In both types of cells, exposure to the highest concentration (0.1 mg / mL) of FDP-NV does not not affect the FBS-induced cell signaling (MAPK Erk1 / 2 phosphorylation) also did not turn on of Caspase 3.
