Polystyrene (PS) and chemically modified compounds in the PS family has long been used in commercial and industrial field. However, poorly understood whether nano-PS or PS microplastic nanoplastic exposure leads to disruption in fundamental cellular functions, such as proliferation, differentiation, and apoptosis. Here, we cultured the three main cell types, including mouse embryonic fibroblasts (MEFs), a mixture of nerve cells isolated from embryonic cortex and cortical astrocytes, and examined the effects of their exposure to nanoplastics PS with a diameter of 100 nm. Despite PS exposure nanoplastic not affect the viability of MEFs or astrocytes, significantly reduce the viability of nerve cells mix. Consistent with the observed effect on cell viability, apoptosis marker levels, cleaved caspase-3, which increased exclusively in nerve cell mixture. To investigate whether cells nanoplastics PS uptake into the cytoplasm, we MEFs and neurons exposed to latex beads PS neon and fluorescence monitored over time. We found that nanoplastics PS stored and accumulated in the cytoplasm with a concentration-dependent manner.
Although astrocytes do not apoptosis after exposure nanoplastics PS, they undergo reactive astrocytosis, with elevated levels of lipocalin-2 and pro-inflammatory cytokines. Therefore, our findings suggest that the susceptibility of cells to the deposition and accumulation nanoplastics PS in the cytoplasm depending on the cell type. Furthermore, based on our data from primary cells derived from mouse brain, we suggest that reactive astrocytosis may contribute to neuronal apoptosis seen in the damaged neurons with nanoplastics PS accumulates in the cells of the body.
Primary bone adult T-cell lymphoma with multiple bone lesions painful and debilitating osteolysis: a case report
Only there are some reports of major adult T-cell lymphoma / leukemia (ATL) in the bone. This is a case report of a patient 75 years initially reported some bone pain associated with osteolytic ATL. Patients developed spontaneous chest / back pain and visited a local hospital. Laboratory tests showed high levels of alkaline phosphatase (ALP), and computed tomography (CT) revealed a bone lesion with osteolysis. Although multiple myeloma was initially suspected, the results of bone marrow aspiration and biopsy of bone inconsistent.
After he was referred to our hospital, mild hypercalcemia (10.4 mg / dL) with normal lower intact parathyroid hormone (PTH) (27 pg / mL), low parathyroid hormone-related protein (PTHrP), and increased 1,25- dihydroxy vitamin D (1,25OH2D) levels (136 pg / mL) to narrow the differential diagnosis lymphomatous and granulomatous disease, and subsequently, high serum soluble IL-2 receptor (3450 U / mL) and the cells of interest are recognized in peripheral blood samples suggesting the involvement of ATL. Finally, reevaluation of iliac bone biopsy samples for histological diagnosis prompted us ATL infiltration in the bone.
Two subsequent courses of chemotherapy in addition to denosumab resulted in a partial metabolic response of interest shown in positron emission fluorine-18-fluorodeoxyglucose tomography / computed tomography (18F-FDG PET / CT). Although very rare, bone involvement ATL can be used for differential diagnosis to local osteolytic bone pain in addition to multiple myeloma and metastatic bone disease.
Fatty-acid-induced FABP5 / HIF-1 reprograms the metabolism of lipids and increase the liver cancer cell proliferation
Hypoxia-inducible factor-1 alpha (HIF-1α) is a transcription factor essential for the survival of cancer cells. Reprogramming of lipid metabolism has emerged as a hallmark of cancer, but the relevance of HIF-1α to this process have remained elusive. In this study, we profile HIF-1α-interacting protein using proteomic analysis and identify the fatty acid binding protein 5 (FABP5) as a partner of the binding of HIF-1α-critical. In hepatocellular carcinoma (HCC) network, both FABP5 and HIF-1α regulated, and their expression levels associated with a poor prognosis. FABP5 increases the activity of HIF-1α by promoting the synthesis of HIF-1α while disturbing FIH / HIF-1α interaction at the same time. oleic-acid treatment activate FABP5 / HIF-1α axis, thus increasing lipid accumulation and proliferation of cells in HCC cells. Our results indicate that the fatty acid-induced upregulation drive FABP5 HCC development through HIF-1-driven lipid metabolism reprogramming.
Behavioral Observation of Epithelial Cells in Nasal Septal Closure During Primary Palate in Mice
Epithelial fusion is very important in palatogenesis and incomplete fusion results in various types of facial cleft, depending on the area that failed to fuse. In palatogenesis mammals, the secondary palatal bilateral fuse in the middle of the face to form the secondary palate. Then, the dorsal side of the secondary palatal shelves fuse with the nasal septum to complete palatogenesis. Importantly, the anterior border of the secondary palatal shelves fuse with the main ceiling, which is located on the border of the anterior and ventral nasal septum. While many studies have investigated the mechanism of fusion between secondary palatal shelves, very little is known about how to touch the ceiling fuse with the primary and secondary palatal shelves. In this study, we investigated the possible behavior of epithelial cells on the surface of the main ceiling using explant culture palatal K14-GFP mice.

Gentaur offers a range of Normal Human Epidermal Keratinocytes (NHEK) from single donors or pooled donors produced at Promo- Cell’s cell culture facility. The cells are isolated from juvenile foreskin or from adult normal human tissue from different locations, e.g. face, breast, abdomen, and thighs. Shortly after isolation, all Gentaur Normal Human Epidermal Keratinocytes are cryopreserved at passage 2 (P2) using Gentaur's proprietary, serum-free freezing medium, Cryo-SFM. Each cryo vial contains more than 500,000 viable cells after thawing. Proliferating cell cultures are made from 500,000 cryopreserved cells that have been thawed and cultured for three days at Gentaur. |

Description: C57BL/6 Mouse Primary Epidermal Keratinocytes from Gentaur are isolated from skin tissue of pathogen-free laboratory mice. C57BL/6 Mouse Primary Epidermal Keratinocytes are grown in a T25 tissue culture flask pre-coated with gelatin-based coating solution for 2 min and incubated in Gentaur's Culture Complete Growth Medium for 3-5 days. Cells are detached from flasks and immediately cryo-preserved in vials. Each vial contains at least 0.5x106 cells per ml and is delivered frozen. Cells can be expanded on a multiwell culture plate ready for experiments under the cell culture conditions specified by Gentaur. The method we use to isolate primary epithelial cells was developed based on a combination of established and our proprietary methods. These cells are pre-coated with EpCAM-1 (CD326) antibody, followed by the application of magnetic beads pre-coated with secondary antibody. |

Observation interval of GFP-labeled epithelial and SEM analysis showed that the extrusion of epithelial appeared in the area in accordance with the fuse region and expanded rostrally on the surface of the nasal septum in the absence of a secondary palatal process. Unlike on the surface of the secondary palate, cell migration and subsequent exposure of mesenchymal autonomous unclear in the nasal septum or palate primer. TUNEL staining revealed that epithelial extrusion undergoing apoptosis. These findings indicate that the extrusion with independent apoptosis begins on the alleged fusion region without contact with the opposing secondary palate.
